Mice have been used in biotechnology medical research since 1894. It is the most common experimental animal in laboratories worldwide. Mice is also been used widely to produce monoclonal primary antibodies. For cancer research pathology study, the researchers often encounter the challenge of staining mouse primary antibody on mouse tissue due to limited choices of primary antibodies other than mouse originated. The major problem is the inability of anti-mouse secondary antibody to distinguish between endogenous mouse immunoglobulins and mouse primary antibody using indirect method. This problem will cause high background staining while the specific staining is then obscured. Endogenous IgGs are mainly distributed in intercellular space and will cause particular problems for antigens localized near cell surface or in extracellular matrix. It is also very difficult for membrane antigens if using mouse primary antibody to detect the membrane antigens on cryostat sections without fixation. The level of background is tissue type-related and varies from sample to sample.
